The epitope of this MAb maps in the C-terminus of PTH, a hormone produced by the parathyroid gland that regulates the concentration of calcium and phosphorus in extracellular fluid. This hormone elevates blood calcium levels by dissolving the salts in bone and preventing their renal excretion.It is produced in the parathyroid gland as an 84 amino acid single chain polypeptide. It can also be secreted as N-terminal truncated fragments or C-terminal fragments after intracellular degradation, as in case of hypercalcemia. Defects in this gene are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. Recognizes a glycoprotein of 110 kDa, identified as CD26. It is an atypical serine protease belonging to the prolyl oligopeptidase family. It is expressed on lymphocyte cells and is upregulated during T-cell activation. CD26 is also expressed on activated B cells and natural killer cells and abundantly on epithelia. MITF (microphthalmia transcription factor) is a basic helix-loop-helix-leucine-zipper (bHLH-Zip) transcription factor that regulates the development and survival of melanocytes and retinal pigment epithelium, and also is involved in transcription of pigmentation enzyme genes such as tyrosinase TRP1 and TRP2. MITF has been shown to be phosphorylated by MAP kinase in response to c-kit activation, resulting in upregulation of MITF transcriptional activity. Mutations of the MITF gene are associated with the autosomal dominant hereditary deafness and pigmentation condition, Waardenburg Syndrome type 2A. Multiple isoforms of MITF exist, including MITF-A, MITF-B, MITF-C, MITF-H, and MITF-M, which differ in the amino-terminal domain and in their expression patterns. Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. Reacts with a protein of 57 kDa, identified as the L1 protein of human papilloma virus type 16 (HPV-16). It is the major capsid protein of HPV-16. Infection with specific types of HPV has been associated with an increased risk of developing cervical neoplasia. HPV types 6 and 11 have been associated with relatively benign diseases such as genital warts but types 16 and 18 are strongly associated with cervical, vaginal, and vulvar malignancies. The metal-responsive element (MRE)-binding transcription factor (MTF-1) stimulates the expression of metallothioneins in response to the exposure of cells to heavy metals. MTF-1 contains six zinc fingers in the DNA binding domain. The phosphorylation of MTF-1 in response to metal exposure appears to play a significant role in the ability of MTF-1 to activate metallothionein transcription. In addition to its role in metallothionein activation, MTF-1 is involved in a post-transcription regulatory complex for ribosomal protein S25. Recognizes a glycoprotein of 75 kDa, identified as low affinity Nerve Growth Factor (NGF) Receptor (p75NGFR) or Neurotrophin Receptor (p75NTR). NGFR is expressed in various neural crest cells and their tumors such as melanocytes, melanomas, neuroblastomas, pheochromocytomas and neurofibromas. Reportedly, anti-NGFR is a reliable marker for desmoplastic and neurotropic melanomas. The protein encoded by this gene is a homeobox-containing developmental regulator associated with liver development. The encoded protein binds to the alpha-fetoprotein (AFP) gene promoter and increases the expression of AFP. This gene is overexpressed in some lung cancers and is linked to poor patient survival, possibly due to its resistance to cisplatin. This gene is aberrantly methylated in pancreatic cancer, deleted in squamous cell lung carcinomas, and acts as a tumor suppressor in esophageal cancer. This antibody recognizes an epitope encoded by exon v3 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. Calretinin (also known as Calbindin 2) is a member of the troponin C superfamily. Proteins in this family have 6 EF-hand domains which bind to calcium. Calretinin is a vitamin D-dependent calcium-binding protein involved in calcium signaling. It is present in subsets of neurons throughout the brain and spinal chord, including sensory ganglia. Antibodies to calretinin are useful in differentiating mesothelioma from adenocarcinomas of the lung. They can also aid in differentiating adrenal cortical neoplasms from pheochromocytomas. Cadherin 17, also known as LI cadherin (for liver-intestine cadherin), is a member of the cadherin family of calcium-dependent adhesion molecules. Cadherins mediate cell-cell interactions via hemophilic binding, and are critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. Cadherin 17 expression is restricted to liver and intestine tissues and is specifically localized to the basolateral domain of hepatocytes and enterocytes.
